The successful candidate will have access to a broad range of technologies to investigate how dysregulation of key oncogenes contribute to colon cancer development, progression and resistance to therapies. Candidates with a good understanding of the mechanistic process that drive cancer progression and/or experience primary/3D culture system to study pathways involved in tumor cell growth are encouraged to apply. This is an ideal position for someone who enjoys the translational aspects of converting basic biology discoveries in cancer development into drug discovery programs and who thrives in a team environment.

Who You Are:

• Qualified candidates will hold a doctoral degree in Cell Biology, Molecular Biology, or Biochemistry and have a successful track record as illustrated by a publication record

• Proficiency in a broad range of biochemical, molecular, and cell biological techniques.

• Key technical skills include:

o Primary organoid engineering using both lentiviral and CRISPR-based approaches.

o Functional genomic screening techniques, both RNAi and CRISPR.

o Cell proliferation assays

o Biochemical techniques including western blotting, co-immunoprecipitation, and in vitro assays using purified components.

About the Lab:

Dr. de Sousa e Melo is currently a group lead and Senior Principal Scientist at Genentech. He leads a discovery research group operating at the interface between technology and biology with specific emphasis on colorectal cancer biology. More recently, his lab has devised a technology that exploits the elevated expression of the cell surface E3 ubiquitin ligases ZNRF3 and RNF43 in colon cancer. More specifically, his team has generated bispecific antibodies that can tether these E3 ubiquitin ligases to a variety of cell surface receptors to promote tumor receptor degradation. This platform that has been dubbed PROTABs for Proteolysis Targeting Antibodies. The in vivo bioavailability and tumor specificity of this approach makes it potentially attractive for the therapeutic management of colon cancer.
